Currently, my job scope in the lab is to assist the microbiologists in the weighing of samples/water for them to conduct
microbial testing on customer samples. I was also assigned to do weekly cleaning (e.g. clean incubators/fridges/waterbaths, etc).
For this week, I had the opportunity to do spread plates. Although i had did it previously during lab sessions in school, it was different as compared to commercial labs like my company. The company purchases sterile spreaders and the microbiologists use it to spread on the agar media in a fast and efficient manner. They dilute the sample and conduct serial dilutions. 1ml of a dilution is spread onto3 plates. As mentioned before, the lab conducts microbial testing on agar media, in duplicates, thus making it a total of 6 plates for each dilution of the sample. When the job is done, these plates are incubated for 48 hours (2 days) in the 37 degrees walk-in incubator in the lab.
I spend most of the time in the afternoon, weighing sterile water for salmonella testing. 1500g of sample is split into 375g for each tub of sterile brilliant green dye water ( 3375g sterile water with 6.75ml brilliant green dye). I got to know from one of my colleagues that this process is known as pre-enrichment, and that it is to be incubated for 20 hours at 37 degrees Celsius. This provides an ideal environment for salmonella to grow. I observed the microbiologists retrieving a certain amount of the “pre-enriched” samples, to test whether is sample is tested positive/negative of salmonella, before disposing these samples.
I hope i get the opportunity and learn more about the different types of microbial tests that is conducted in a commercial lab, and also be involved in media preparation, etc.
SIP day 3
Learnt how to autoclave water. Labelling as usual.
Learnt how to weigh product samples :) 10g!
Then continued to weigh water for sal. testing.
then went to the washing area and learnt how to wash the dirty stuffs like test tubes, bottles this kind.
Also had to go drag the big green trashbin to throw all the biohazard trash.
Learnt where to go get RO water too.
Learnt how to clean the waterbath too, and mop/sweep the floor.
SIP day 4
Jiaxi wasn’t here.
Continued to fill up water for autoclave
and then weigh water again for salmonella testing.
everyone had a meeting with boss today. he mentioned some stuffs that i dun understand. i diam diam.
Mopped the floor again. guess im going to do this everyday.
SIP day 5
Autoclave water. and then went through theory of weighing environmental samples with JX. With Anna showing me how to do :)
Then weigh water again. For long hours. I keep wasting water! Man! I feel useless =.=
Brought the bottles back and “cho-lo” i mean careless me always knock corners with the trolley zzz damn cramp coz got some Caucasian came and put a laptop and some equipments there.
Mopped the floor after 5+pm and then went home :)
Didn’t go for company dinner coz i felt tired.
Okay now thinking of what to write in the journal. I was chatting with OYN on msn just now and he gave me good ideas. lol.
- I learnt more about the SOPs, and their general practices in the microbiology lab of a food testing company.
- I also learnt how to handle the different equipments in the lab (e.g. autoclave machine, waterbath, etc), with the emphasis on how to sanitise and clean them. It is important that the equipments are sanitised/clean before use, as it also might be a possible contaminant. Viraclean/ 70% alcohol/ trigene is normally used in these cases.
- I learnt that sanitation is extremely important in the microbiology lab as possible contaminants may be present (from the air, from body, etc), and we should always take extra care to sanitise before conducting any form of work (e.g. weighing samples), so as to make sure that the microbial readings obtained are as accurate as possible.
- I also learnt on how to weigh environmental and product samples, which the experienced microbiologists would conduct serial dilutions and microbial testing on these samples to detect the presence of salmonella/listeria or even general micro like B.cereus, S.aureus, etc.
First Day
My supervisor was absent o_o
Was a blur sotong today lol. Entered the lift (when the lift stated need to have a pass to go up!) and someone tap their card. LOL. what’s the purpose of cards -.-
Btw because of this stupid pass system i had to trouble Alan to open the door for me twice (then he gave me my card). haha. Now got card liao feel like a staff! (:
First day I had to tear the stickers of test tubes meant for disposal then stack them together for them to autoclave and throw.
Then have to transfer the coloured liquid samples to the Pasteur cupboard for autoclaving :) carried the “microbial load”! Heavy! haha!
Then had to sanitise the white and red bins with trigene and put them into incubator to dry.
Then have to tear off stickers again and such.
Day 2
Early in the morning we autoclaved water :)
I filled up the water and load into autoclave.
Then had to do labelling of MYP and BPA media.
But most of the whole day i had to weigh the water for salmonella testing.
Used the pipette gun! Damn cool! Haha so convenient! Sprayed brilliant blue dye in the sterilised water :)
But had to carry those 5L bottles to pour 3+L of water for each tub….I must have did hundreds…my shoulders aching like crazy.
Then it was raining today and silly me volunteered to do extra work after 5.30pm. lol.
Loaded the sterilised water and stuffs from the pasteur cupboard to the trolley. and did labelling. Yeah labelling’s fun! Ahaha.
Then went home riding bus 80. damn pain my leg. coz had to stand till hougang till lots of ppl get off then i can sit =.=

Today have to listen to sensei’s ideas for advertising and blah blah blah.
Then sensei went to sleep when we did self-training.
We just mainly do stretching and conditioning today o_o
Went for lunch at 2pm without sensei
Ate at Four Seasons, a small taiwan restaurant.
I ate a Pork Cutlet Udon Soup and A pearl milk tea! Yum yum :D
Then new committee interview there also coz simon had to go off earlier o_o
Then went to buy steamboat ingredients with Red, Wind, Justin, Victor, Leslie, Rachel, Velda, Jamie, Celesta, and Joey. Walked to parkway parade Giant through Roxy square. Never knew of this good shortcut o_o Spent $100+ (coz also buy sushi platter and yu sheng).
Then went back. I ran to 7-eleven buy ice. Only in 5kg pack. lol. Costs $5.20. Expensive -.-
Then prepared for steamboat and yu sheng :)
Played sparring kendo with the sponge sticks with Victor XD
My specs keep drop off lol. Hit damn hard.
